چکیده
Abstract Downstream signalling involving adenylyl cyclases (ACs) and kinases is a key component of several processes in plants including cell division, growth, response to stress. ACs are enzymes that generate the second messenger molecule, 3′,5′-cyclic adenosine monophosphate (cAMP) from 5′-adenosine triphosphate (ATP) while catalyze addition phosphate group other molecules (trans-phosphorylation) or themselves (auto-phosphorylation). Apparently, there has been an expanded record various identified plants, no plant molecule date shown possess both AC kinase activities/functions with such having characteristic cross-talking interactions. Therefore, endeavor find we searched amino acid sequence known Arabidopsis AC, pentatricopeptide repeat (AtPPR) protein, found kinase-specific signature (KSSS), which speculated be working synergy center this protein during downstream signalling. So, order test if additional catalytically active perhaps also some interactions center, cloned, expressed, affinity purified truncated version AtPPR, harboring KSSS centers (AtPPR-AC/K). When tested vitro, recombinant AtPPR-AC/K showed Mn 2+ -dependent activity positively enhanced by Ca HCO 3 − trans-/auto-phosphorylation capable utilizing ATP GTP as substrates specific serine, threonine, tyrosine acids target residues. In addition, was reduced cAMP at same time, it totally shut down . This thus qualified molecular switches modulators, regulating AtPPR functions through between activities its two domains. Our work, therefore, essentially established first member new class moonlighting proteins have themselves, conceivably presenting ideal candidate for further explorations improve particularly agricultural crops.

An Arabidopsis pentatricopeptide repeat protein, SUPPRESSOR OF VARIEGATION7, is required for FtsH-mediated chloroplast biogenesis.
The Arabidopsis (Arabidopsis thaliana) yellow variegated2 (var2) mutant has green- and white-sectored leaves due to loss of VAR2, a subunit of the chloroplast FtsH protease/chaperone complex. Suppressor screens are a valuable tool to gain insight into VAR2 function and the mechanism of var2 variegation. Here, we report the molecular characterization of 004-003, a line in which var2 variegation ...
متن کاملAn Arabidopsis dual-localized pentatricopeptide repeat protein interacts with nuclear proteins involved in gene expression regulation.
Following the endosymbiotic acquisition of mitochondria by eukaryotic cells, most of the genes in this organelle were transferred to the nucleus. To maintain mitochondrial biogenesis and function, nuclear and mitochondrial genomes require regulated and coordinated expression. In plant organelles, nuclear-encoded proteins targeted to the organelles control posttranscriptional and posttranslation...

UNLABELLED Little is known about intracellular signaling mechanisms that persistently excite neurons in pain pathways. Persistent spontaneous activity (SA) generated in the cell bodies of primary nociceptors within dorsal root ganglia (DRG) has been found to make major contributions to chronic pain in a rat model of spinal cord injury (SCI) (Bedi et al., 2010; Yang et al., 2014). The occurrence...
متن کاملFormation of the Arabidopsis pentatricopeptide repeat family.
In Arabidopsis (Arabidopsis thaliana) the 466 pentatricopeptide repeat (PPR) proteins are putative RNA-binding proteins with essential roles in organelles. Roughly half of the PPR proteins form the plant combinatorial and modular protein (PCMP) subfamily, which is land-plant specific. PCMPs exhibit a large and variable tandem repeat of a standard pattern of three PPR variant motifs. The associa...
متن کاملDifferential activation of adenylyl cyclase by protein kinase C isoenzymes.
Cyclic AMP production within cells is altered upon protein kinase C (PKC) activation; however, whether PKC directly modulates adenylyl cyclase (AC) catalytic activity has been controversial. Molecular studies have elucidated the existence of multiple PKC isoenzymes although the functional role of this diversity is not clear. Using purified PKC and AC isoenzymes, we demonstrate that PKC zeta dir...
